What kind of connection (infra red, serial, USB, Ethernet)? Ethernet
won't work until you connect via some other method _and_ set up a
partnership. For the others, make sure you've set the handheld to use
that method when connecting, and make sure you've set ActiveSync on
the "desktop" (other end of ActiveSync) to allow that type.

Some handhelds have an option to connect manually (user explicitly
starts ActiveSync on handheld) or automatically (ActiveSync starts as
soon as physical connection is made). If both are available, try both.
Also, try letting ActiveSync on the "desktop" initiate the connection
(File->Get Connected). I've had a couple of handhelds that needed
extra coaxing: start ActiveSync manually on the handheld _and_ start
    Get Connected on the "desktop".
