Did you download the entire SP4 package or are you attempting to install it from within Internet Explorer via Windows Update ?

If you're downloaded the entire package and are receiving error 0x0643
along with a message about IE running, and you did not open IE, then as Robert has already pointed out, there's a strong likelihood that the system is infected with malware.

Please let us know which method you have chosen to install SP4 ...
either via Windows Update with IE open, or, by downloading the entire SP4 package. OR, if it was by another method.
