Yes, it's a plain text file.
I don't know what file you mean with "the existing .inf file".
You shouldn't have an existing ntprintsubs.inf file, unless someone
else already tried to define substitute printer drivers.

How to create the .inf file, use normal notepad and save it as
ntprintsubs.inf ?
I can't modify the exiting .inf file, when i open the file, is
garbage.

You can replace the printer name on the right side ("HP
LaserJet Series II" in the example above) with any printer
driver name which exists on your server.
Note that this file is case sensitive, so "hp LaserJet" (as you
wrote in several places) is *not* equal to "HP LaserJet" and
won't be recognized by Windows!
Follow the instructions in KB 239088 to create the registry
keys to point to the ntprintsubs.inf file.
