From your post, I understand that when attempting to backup to a tape,following error messages are received:
1. Write protected media.
2. Error: C: is not a valid drive, or you do not have access.
If I have misunderstood your concern, please feel free to correct me.
This issue can be caused by several factors. It's hard for us to determine
the root cause based on current information. The troubleshooting can be
complex and time-consuming.
If the issue is urgent to your business, it is recommended that you contact
Microsoft Customer Support Services (CSS) via telephone so that a dedicated
Support Professional can assist you recover the server in a more efficient
manner. Please be advised that contacting phone support will be a charged
call.

For this problem, in order to narrow down the cause and resolve the issue
as soon as possible, I suggest you perform following test:
1. In order to make sure the backup system on the SBS server is functioning
correctly, I would suggest we schedule a full server backup with the SBS
backup wizard in the server management console. Please set the backup
destination to one of your local hard drive. Please let me know the result.
2. If it is convenient, please try to backup the system to a new tape and
check if it works on the new tape.
3. Right-click "My Computer", and then click "Manage", expand "Storage",
then expand "Removable Storage". Click Libraries, check if the tape drive
is connected well.
4. If it is possible, please connect the tape drive to another computer and
check if NTbackup works.
5. Please backup only one test file from the C drive to the tape. What's
the result? Please send me following backup log:
C:\Documents and Settings\SBS Backup User\Local Settings\Application
Data\Microsoft\Windows NT\NTBackup\data\backup09.log
6. Please send the MPS report.
a. Visit

15706/MPSRPT_SETUPPerf.EXE to download the file.
b. Run the MPSRPT_SETUPPERF.EXE on the server box.
c. Wait for 10~15 minutes.
d. Open Windows explorer, navigate to
%SYSTEMROOT%\MPSReports\Setup\Reports\cab\
e. Send the .cab file directly to me at v-mzhuan@xxxxxxxxxxxxx
