the smtp service is running and see the connect attemp from telnet, check
your connection setting on the smtp virtual server (security ipaddress
range.....) it seems the connection is closing the moment you open it. stop
the default and create a new virual smtp server, start it and see, if you do
not stop the default you will get an error message the port is in use.

this line jumped out at mean,
where do you see the connection attemp?
event log or SMTP log?

I can't find any errors when I stop/start the smtp service. In the event
logs there are only notifications that SMTP service stopped and started, and
SMTP service instance 1 stopped and started.
I thought this might be odd, so I checked on my local server and when the
service is restarted, two instances get restarted as well.
There are no errors in the SMTP log files either, I only see the telnet
connection attept that I initiated from the server itself.
I have installed no other software on this server other than the antivirus
and the UPS monitoring software - both of which I have disabled.
I'll try a virus scan, and we'll see how it goes. At this point I'm not sure
what else it could be. Thanks again for all your assistance!

Clear all logs(save them)
then restart smtp. and then look for errors in the event system. Do not
forget the IIS smtp logs also. these logs will tell.
I almost willing to bet you have another smtp software running. Did you
install any spam scrubbing software.
I know you have other sbs box setup the same way. but do not let this cloud
you trouble shooting process. I can 5 idental box with 5 different issue. so
you can use as ref. but thats it.
RUN AN ONLINE AV SCAN>TRENDMICRO.COM OR BITDEFENDER.COM an SMP spamming
virus code have attached the box.

This particualr box is standard edition with one nic.
I tried changing the smtp listening IP address to the local interal IP
address, and restarted the SMTP service. Still no response when attempting to
telnet from a PC on the network.
As far as the Anti-Virus, I'm using McAfee Corporate version 8, same
software I use on all my client servers, including my own - this has no
firewall built in. I've disabled it anyways, just to be sure.
I'm thinking about using the change IP address tool to reset things, using
the same address. Let me know if anyone thinks this is a good idea.

Check in system manager under server name protocols SMTP, change the SMTP
lissening port to internal NIC, restart smtp and telnet into it from the
server or internal workstations. if it answer then go back and change it to
the WAN nic and do the same test.(use IP address not domain name as it is
another test.) note which nic answers.

Do you have ISA install, if so turn off and run test.
ISA manager service right click stop firewall and proxy services or use the
service manage mmc to stop service also
Look at the AV, If it has any firewall stuff even turned off it can keep
smtp blocked!

I'm running into a weird problem with email on an SBS 2003 SP1 box.
All email is getting bounced back, and I can't seem to figure out why.
Email is setup to be delivered directly to the server through the wizard,
all DNS records are setup properly for this - when I use an online MX tool
the correct IP of the server is returned.
From the server itself, I run an online port scan tool, and it reports that
port 25 is closed. I have checked the DLINK firewall, and the port is
forwarded correctly, and in the firewall log I see no requests to port 25
being rejected during the port scan.
The only thing I can think of is that the smtp server on the box is not
listening. I've disabled the anti-virus (McAfee Enterprise), and still
nothing.
This is very strange because I have numerous SBS boxes setup exactly the
same way, with no issues.
I'm hoping that someone out there can shed some light on this, because I
seem to be running out of options. Have I missed anything?
