A defrag won't clear any space - that's not what a defrag does. It simply puts file fragments back together in contiguous space. There can be lots of things taking up space on the drive, but one of the obvious ones is Volume Shadow copies. If you have that turned on for D:, then I'd re-point it to another drive for its storage.
Moving the exchange files is fairly trivial, by the way, so if you have an alternative drive to put them on that has plenty of space, I'd start by doing that.

Interesting view on stopping Exchange before using the Windows defrag, as I googled to another post which said leave it running as exchange will lock the files and keep the der*** from interfering with the databases.
My basic problem is the D: (exchange) drive getting full. We had a warning a week or two back, and I was able to free some space as a temp measure, but I can't work out where the capacity is being consumed!!
D: is 18.5GB with presently 2.24GB free. Total used 16.3GB
But if I look at the individual root folders on D: I get:-
Client Apps - 1.02GB
Exchsrvr - 6.34GB
Fax - 0
Recycler - 1.73MB
Service Packs - 4.89MB
System Volume Information - 0
Tickets.com - 577MB
Nowhere near 16.3GB
So where is it all going?
Hence a der*** to try and free some space?
Any pointers appreciated.

I have run Eseutil /d on the D:\exchange database with no problems.
The generic windows Disk defragmenter when analysing the D: partition still advises defragmentation. Is this safe to do, with the exchange MDBDATA data contained on it?
Presumably it will be safe to defrag C: and E: if advised by the Disk Defragmenter?
I have backups, and also exmerged exchange to pst files, just in case!!
