A couple of observations on your question,
1 as you discovered adding up the sizes of the mailboxes does not equal the
total size of the store. Make sure that you are not scanning the exchange
databases with your antivirus software. That can cause the store to become
corrupt and give you the file size issues you had. Also remember that when
you delete items in the mailboxes the size of the store will not be reduced
until online maintenance has run, the deleted mail retension has expired
and an offile defrag of the database has completed
2 In reference to the backup size. If your tape drive does compression to
reach its full capacity, that is usually based on an average 50%
compression ratio. The fact is alot of files don't compress that much. The
type of data that is being backed up should influence your calculation of
how much one of your backup tapes will hold.
