I've run into a problem before, made changes, and everything appeared
to work for awhile...then a couple of hours later I had DNS problems
again.

First, make sure all internal clients use the internal DNS server, only.
Even if you have only one internal DNS server, it is the only one that can
resolve the local addresses. No external DNS servers allowed in any
position.
Then, run ipconfig /flushdns and try accessing the site again.
