As long as you're pointing to the internal DNS servers on all interfaces...

1. Are any services disabled such as the DHCP Client service? (it's
required).

2. Is the firewall service enabled? (disable it).

3. Did you alter any C: drive permissions and installed a recent hotfix?
That will kill network connectivity.

4. Is the File and Print services enabled?

5. Microsoft Client Services enabled?

6. Is DNS listening on the private LAN interface?

7. More than one NIC?

8. Updates allowed on the zone?

9. "Register this connection" checkbox enabled in NIC properties, IP
properties, Advanced, DNS tab?

10. Primary DNS suffix matches the zone name in DNS and the AD domain name?

11. Was Zone Alarm ever installed on these machines?

12. Any Event log errors?

13. Spyware or somethng else such as DotNetDns installed on it?
