The Windows Server 2003 R2 schema upgrades are required for R2 features.
See http://technet2.microsoft.com/windowsserver/en/library/84445c1b-a418-4a09-a50c-5f3258cfc5b51033.mspx?mfr=true, which lists three R2 enhancements that require the R2 schema upgrade:
1. DFS Replication service
2. Identity Management for Unix
3. Deploy Printers functionality of the Print Management Console
This document also says:
"You must update the schema before you upgrade an existing domain controller to Windows Server 2003 R2 or before you add the Active Directory domain controller role on a computer running Windows Server 2003 R2 by using the Active Directory Installation Wizard (Dcpromo.exe). You must extend the schema before you perform either of these actions for the first time in an existing forest. "
The page at http://technet2.microsoft.com/windowsserver/en/library/84445c1b-a418-4a09-a50c-5f3258cfc5b51033.mspx?mfr=true says essentially the same thing. If you want to add a Windows Server 2003 R2 server as a Domain Controller to an existing forest, you have to update the schema first:
"In forests that contain the Windows 2000 or Windows Server 2003 schema, you must use the Windows Server 2003 R2 Adprep tool to add the Windows Server 2003 R2 schema additions to Active Directory before you deploy the following features or server roles:
. Distributed File System (DFS) Replication Service
. Domain controllers running Windows Server 2003 R2
. Identity Management for UNIX
. The Deployed Printers functionality of the Print Management console"

You do not need to update the schema, unless you are adding Exchange.

In a domain, I have until now only one DC running on Windows2003 SP1.
I want to add a second DC running on Windows2003 R2 and let run AD replication between both.
Should I run an Active Directory Schema Update on the first server before promoting the new one?
Is running the Active Directory Schema Update on a production machine something that does not disturb daily operations? (no reboot, no change on clients...)?
There are a lot of articles on technet about this. Could you give me the link(s) which are suitable? (write access to schema?).
