No and no.

Do I need to upgrade the master DC to R2, if applicable?

You need to run adprep from the second disk of the R2 set on the schema
master for your domain. You need to install the R2 features on the servers
you want to *use* the updated R2 features. Example, if you are going to use
the new DFSR that comes with R2 and replicate data back to the corporate
office from a branch office server. You will need to install R2 on each of
the servers you want to participate in the replication group. You would
update the master DC to R2 if you want that master DC to use any of the
updated R2 features.

I have a Win2k3 Domain Enterprise with SP1(non-R2). I have 20 DCs that

non-R2 and currently with SP1. I have 20 new servers that I am planning
on
building with Win2k3 R2 SP2 and promoting to DCs. I would like to know
if
there are any caveats that I should prep for since the new DCs will
have
newer OS release than the master DC hosing the FSMO.
