I would check the following:
1> Make sure that the Domain Administrators group has rights to the GPO.
2> Make sure that the (PDC Emulator) FSMO role is funcitoning.

When I try to access my Domain Controller Security Policies I get:
Failed to open Group Policy Object. You may not have appropriate rights.
The system cannot find the path specified.
Nothing has changed in the domain, but my exchange server says it cannot
connect to domain controller. Uh....its the same server. I do register
Event ID 1000
I have checked DNS Services as well as Sysvol security settings. Seems
OK.
