Avoiding using large numbers of groups

We're moving a large number (over 500) of shared folders from Novell to ADS.
The folders are shared currently to groups of from 2 to 400 users and we'd
been using Groups on the Novell side.
I understand that in ADS Groups tend to slow things down, so, what's the
"best practice" to setting up access to shared folders WITHOUT using Groups
(like being members of an OU?)--or, is that possible?
