Not sure how familiar you are with group policy administration, but there is a setting in AD that allows you to set precidence for different policies. If you created a new policy that either overrides settings in your default/parent policy, or needs to be applied later, you will need to change the precidence of the policy in order to allow it to be applied after the default policy.

I'm not exaclty sure what you did in terms of your OU structure, but if you have a multi-tier organization that will require different policies for different groups of users, it may be advantageous to create a policy for each group, and break the groups into several OUs, and then apply policy to each OU on a case by case basis.
