Can anyone tell me which registry value sets the "Run unsigned Content" to
Enable/Run in sandbox/Prompt?

To get there:
Internet Options | Security | Trusted Sites | Custom Settings | Java
Custom Settings | Edit Permissions | Unsigned Content | Run Unsigned
Content | Enable

Use RegMon and filter on iexplore*SetValue and find out.
(assuming you are using IE to display and use the Internet Security
Properties dialog, e.g. by doubleclicking on the zone icon in IE's status bar.)

Note: RegMon is freeware from SysInternals.

Good luck
