If you have roaming profiles so that the TSs can access the profile, then yes, you need to move the redirected folders back to the profile, in order to be able to access it from everywhere.
On the second tab of the GP settings for folder redirection, I can set the behavior for the event that the policy is removed. Now, how do I properly remove that policy to copy the data from the dedicated share to the profiles? Simply setting the Setting in the policy to 'unconfigured' does not seem to work.
You need to redirect the folders to another place - simply deleting isn't enough. Use the share you have the profiles on or a local place.
