If you really have user accounts in the OU you just applied the GP to (no groups etc.) then the policy should apply. Anyway, as it seems not to work, I have a few things for you to check:

I assume that the computers are correctly joined to the domain and DNS is working well (the clients have configured your DC as the first DNS server and can query it...).

- Log on as a user that should get the policy applied and see, if your policy shows up on "gpresult" (Start->Run).
You can also access these information by clicking Start->"Help and Support" -> Click "System Information" at "Tools - Locate tools like System Information..." -> Click "Advanced
System Information" at the left panel under "Tools - Help and Support
Center Tools" -> "View Group Policy Settings applied".

- Are there any errors shown in the event log?

- Do other policies work with that OU and these users? If so, try to assign the following GP as well (notice, that this is a computer configuration - you will need to apply this to a OU containing computers..)
CompConf\AdmTemp\System\Group Policy\ - "Internet Explorer Maintenance
policy processing" and there: "Process even if the Group Policy objects
have not changed".
