Applying Group Policies

I am learning how to apply group policies. On a Windows Server 2003 I
created a GPO to limit certain things such as access to the run menu...etc.
The GPO is linked to the OU however the changes are not being reflected on
the client workstations. I am not sure what I am missing.

Also I do not have a clear understanding as to how "No Override" and "Block
Policy Inheritance Works"

Any help would be greatly appreciated.
