Restore partition defrag'

After 3yrs 4.9 Gb Fat32 partition D of my 200 Gb HD is badly fragmented but there is not sufficient contiguous free space to allow defrag of some larger files. There is plenty of space on partition C.
Could I temporarily move partion D's contents to NTFS C, defrag then rewrite to D?
