I installed TweakUI on my work laptop (XP Pro) just to change one
setting: General > Focus > Prevent applications from stealing
focus.
Unfortunately, every time I log on, the setting is lost. Newly
created windows are still capable of interfering with what I am
doing, and TweakUI shows the box unchecked again.
I log on to a VPN, not a local user account, so perhaps my
settings are being downloaded from the network every time? (I
don't know how this works and could be wrong about it, but on my
non-work computer, with a normal user account, TweakUI settings
don't get lost.)
What can I do to make this setting persist?
It sounds like you might have some Domain Policies that changes this
back. What you might try is to make the changes in the registry
instead of using TweakUI. Instructions for doing it are here:

When you have finished making the registry change, click once on the
registry name (ForegroundLockTimeout) to select it, then click on:
Edit->Permissions
Then for every group or user name that is listed, change it so that all
users have only Read access to this registry entry. This *should*
prevent it from changing back. Even _you_ should not be able to change
it back unless you un-do the security permissions.
