I read somewhere that if the pagefile is too small the defragmenter might not work properly. The suggestion, which apparently works for many, was to set the pagefile to a static size of 2X RAM (twice the size of installed RAM). Easy enough to try, see what happens.
Other than that the only thing I can suggest would be to run an sfc /scannow on the machine.

Look in the %systemroot%\system32\dllcache folder for regsvr32.exe and copy it to the system32 folder and try again. Something may have gotten skewed with your Windows installation, the regsvr32 file should be in the system32 folder. Did you eliminate the possibility of virus or other malware on your system?
regsvr32.exe is in the system 32 folder,could not find dllcache folder,ran spybot,AD Aware, AVG.did chkdsk after reboot, everything comes up ok, 47% free space on disk, deleted temporary internet files, tried reinstlling defragmenter from WINDOWS\ inf folder. So far back to square one defrag will not run. Also tried a restore point back 7 days.
Do you have enough free space on the disk for the defrag tool to work? Did you do a disk cleanup and get rid of temporary internet files? Sometimes these files can cause the defragmenter to have hiccups.
If all else fails try reinstalling the defragmenter. Look in the WINDOWS\INF folder for dfrg.inf right click on it and select install.

Try registering the defragmenter dll's. From a command prompt issue these three commands, pressing <enter> after each command (line):
cd %systemroot%\system32
regsvr32 dfrgsnap.dll
regsvr32 dfrgui.dll
both commands come back with "not recognized as an internal or external command operable program or batch file"
