If (and only if) the hardware in both systems is identical you can do this. Otherwise as others have said, a repair install will be needed.
My question to you is why don't you just set your wife's PC up to share files/printers? It's not that difficult to do and doesn't require swapping drives around. In other words, I think you're approaching it from the wrong direction.
