modifying sharepoint

Hi,
We have SharePoint as running on SBS2003 (companyweb)
which we use for out intranet. We set everything up last year fine.
Now, we want to change companyweb by creating a new list.
However whatever we do, whatever the account on whatever the machine
we get an access denied message after the windows authentication
window pops up. This includes the admin account on the server.
We have no idea what is going on. The user administration part of
SharePoint is less then satisfactory to use.
I would be grateful for any pointers as it is driving us nuts
