Maybe it's because you have not enabled the mail profile from the properties of SQL Server Agent? Check the SQL Server Agent's properties \ Alert System.
Also, what's the name of the mail profile? And what's your SQL Server version? (Has SP2 been applied?)

I have setup a new installation of SQL Server 2005, parallel to our
production server. SQL Mail works fine on the production server. On the new
server, configured as identically as we know how, we can get the "test"
function to work.
However, in SQL Agent, we encounter the following errors:
"Unable to start mail session (reason...mail configuration information could
not be read from the database --> System.Data.SqlClient.SqlException: profile
name is not valid"
So why can we run a test successfully if the profile name is not valid?
Where to we begin to look for clues?
