configure multiple instance in enterprise manager

I have created two instances of MS SQL on the Database server. From a client PC I am using Enterprise manager. I have configured and added my first default instance. When I tried to add the new instance using new sql server registration, it gives me SQL server does not exist or access denied Connection open. I can create a new SQL group but it has no items in it.
Your solutions are greatly appreciated.
