Upgrading from MSDE to SQL Server is deceptively simple: Run the SQL Server
installation. You will be asked if you want to upgrade your existing
instance or install a new one. Choose to upgrade. Afterwards re-install
SP3a. None of your application databases or security settings will be
changed.
If you are needing to migrate from MSDE to SQL Server (move the database
from one platform to another) then you will need to provide instructions for
detaching and re-attaching your database and setting up the logins and
security settings on the new system.
