Yes, I know it is a Security issue but I have no idea where.
The Share Permissions are set to Everyone Full Control
The NTFS Permissions are set to:
Guest Read & Execute
List Folder Contents
Read
Users: Read & Execute
List Folder Contents
Read
In the IIS Setting we have select under Authentication Methods.
Enable anonymous Access
Integrated Windows authentication (This Point was not selected when I start
my Troubleshooting, I have select it later. Can that be the Problem)

We have a SMS Installation with 10 Sec. Sites
In one secondary Site the Clients cannot Access the DP.
In the DataTranferService log on the Client I get the following Error
message

Error sending DAV request. HTTP code 401, status
