Have you checked the ReadMe? I think you may be looking for the third
section "Set Up the Script"?

Set Up an HTML Signature

Go to Tools/Signatures in Entourage, and click the New button. Name it
"Auto-Reply HTML" or something similar. Click the abab button at the left
end of the message composition toolbar to enable HTML. Type your full HTML
auto-reply, including a signature at bottom, using all the color and rich
text devices available. You can even insert a graphic wherever you wish in
the message (although it won't always be appreciated by recipients on
pay-per-second dialup connections, of course). And animated graphics don't
work, of course, so don't bother with those. When ready, click Save. You can
always come back at any time to modify this auto-reply.

Set Up a Rule

Go to Tools/Rules/POP or IMAP, or Hotmail, as appropriate in Entourage, and
click the New button. Name it "Auto-Reply HTML" or something similar. Set
your criteria for which messages should be acted upon. (If you were planning
to apply it to "All Messages" for a period of time, instead make the
criterion "Is not from a mailing list" ‹ do not torment all the mailing list
readers with this auto-reply to every list message!). Set the Action to Run
AppleScript, click the ScriptŠ button, and navigate to Auto-Reply in HTML
script in the Entourage Script Menu Items folder.

Set Up the Script

Go to the Script menu in Entourage and select Auto-Reply in HTML . It will
present you with a list of the names of all your signatures. Select
"Auto-Reply HTML", or whatever you have called it.

Now you're all set up. You can always modify the text of the auto-reply
"signature". [If you should ever want to replace it entirely, delete it from
the list of signatures in Tools/Signatures, make a new one with any name,
then run the script once from the Script menu to set it up again.)

Operation

Operation is automatic if your "Auto-Reply HTML" rule is Enabled. You can
disable it by simply unchecking its Enabled checkbox in the list of Rules or
within its own Rule window. Enable it again by checking the box.

You can test it out by selecting a message you received (best if it's a
message you sent to yourself), go to Message menu/Apply Rules/Auto-Reply
HTML. You'll see the auto-reply quickly form itself in the front and get
sent off. That's exactly how it will work automatically from the rule too.
