In Explorer, right-click on the folder -> Properties -> Security
Give IUSR_<Machinename> NTFS Read (R) permissions.

What you are doing is configuring IIS to use a particular account as the
"anonymous user account". This account then needs to be given NTFS
permissions to the folders/files in question. If you change the anonymous
user account in the IIS manager, then you need to use Explorer to change the
NTFS permissions on the files in question so that the new account also has
access.
