If you chosed to run IIS SMTP on your computer, then you could use your computer to send out the email
from your website because SMTP would be running.
I highly encourage and recommend as an Systems Administrator of 7 years to please read all you can
about securing SMTP on IIS if you decide to use it. Here is one article I came across using
Google.com:

In other words, you do not want your computer left open as a possible spam relayer. This happens with
many people who turn on SMTP and don't do a thing to secure it.
Looking through PHPNuke's site, I have found a thread where several people recommending using other
Mail Transfer Agents (or MTAs or commonly known as Mail Servers). Check their recommendations for
having your PHPNuke sending out emails:

Other than that, you would have to check the settings for PHPNuke (either manually or within the Web
Console) to use an external webserver. I am only familiar with running PHPNuke with an internal SMTP
service running.
