What do you mean? Are you running a site-to-site VPN with Cisco at one end
and ISA at the other end of the tunnel?

With ISA2000 you don't configure anything in particular, you just have to
make sure that routing between the networks involved is properly configured.

What client? All you mentioned so far was a Cisco router used as VPN server.
