Exchange cannot do this by default. But it can be done. You will either
need to write or to purchase an SMTP sink that will integrate with SMTP to
provide this functionality. The downside is that you will want to dedicate
a box to doing outbound SMTP and even then it will slow it down a bit.

If you are looking to replace end users signatures this will be very hard as
a signature is just part of the body of the message and is not separated
from the message in any way manner or form. It is easy to add a
"disclaimer" or other canned text to the end of all messages.

In this case it will most likely be easier, faster, and cheaper to just have
all of your users change their signature as appropriate.
