If you're taking an offline backup, which shouldn't be routinely necessary,
you will have to stop the Information Store service, and that will
disconnect all users.
You should take an online backup of the Information Store using NTBACKUP or
a third-party product with their Exchange Agent, and when doing that there's
absolutely no need to disconnect Outlook users. If you take a file-based
backup of other things on the server exclude all the EXCHSRVR\*DATA
directories.
