What do your SMTP protocol logs tell you is going on at that time? Do
you see commands being sent and rejected? What does a network monitor
show at that time?

I have since enabled logging on exchange and noticed that this #5.5.0 NDR is
occuring on average once or twice a day, at different times from different
senders.

Also, twice when i have received this 5.5.0 personally and i looked on the
queue straight away and there was a large number of emails in the queue, when
usualy there is none (as it should pass these to the smarthost for queueing),
but one other time when i got this 5.5.0 error there was no messages in the
exchange queue, so im not sure if there is any relation to these two problems.

When there is a large number of messages in the exchange queue it seems to
be "blocked" and i need to change the connector to use DNS rather that
smarthosting then all the emails send, then when i change back to "smarthost"
for the connector everything seems to work again.... again im not sure if
this is related to the 5.5.0 error or if it is a seperate issue.

That's not an expected behavior. Have you verified that the switch(es)
and your NIC are both using the same connection speed and duplex?

What release and service pack of the Windows O/S are you using? Have
you run the Exchange Best Practices Analyzer on the Exchange server?
Any chance you're using the Scalable Network Perfomance stuff in W2K3?
If you are, either turn it off or get the update for it to make it
work.
