It's generally not necessary to restart anything for MRM policies to kick in.

So I launch Outlook this morning, and voila -- I see that suddenly the MRM
policy has taken effect. I check another test account I had applied the
policy to, and the same was true for it.
What changed? Well, last night I made a configuration change to the server
that hosts Exchange and had to reboot it. Looking in the MRM log files, I see
that policy moved the Junk E-mail messages to Deleted Items just a few
minutes after the server came back up after the reboot.
I'm guessing that cycling one or more of the services is what kick-started
MRM. This should not be necessary, though--or if it is, it should be
documented somewhere.

The policy is definitely configured to manage the Junk E-mail managed
default folder.
The Junk E-mail managed default folder definitely has Managed Content
Settings under it.
Looking at the properties of those settings, it definitely says 1 day for
the length of retention period. Retention period starts is definitely "When
item is moved to the folder." Action to take is definitely "Move to the
Deleted Items folder."
I'll try a second mailbox, and will change "retention period starts" to
"when delivered."
In the mean time, I've got a question posted to
microsoft.public.exchange.connectivity that has your name written all over
it...
