Yes it will work. Exchange will block emails from hosts in the Deny list and
"whitelist" emails from hosts in the Accept list. All the others are not
affected by Connection Filtering.
I suggest you to check the following for a known issue with connection
filtering:

By the way the double qutoes around whitelist are because of this:

Hello.
We are using Exchange Server 2003 SP2 on Windows Server 2003 enviroment.
I enabled IMF, Recipient Filtering, Sender Filtering at this moment.
I would like to enable Connection filtering as well tho, I have one quick
question.
If I don't add any Block List Service configuration and just use Global
Accept/Deny
List configuration, in other words, just use Connection filtering as
Global Accept/Deny List,
is it going to work? or since there is no server verifying w/ all e-mail
coming in, Exchange
is going to block all e-mail? or just pass thru all mail except matched IP
addresses
listed under Global Accept/Deny list?
