Why don't you try this:
Send a message to the user that is under 10240K. Does it work?

I'd guess that the problem is your mailbox store defaults. You could turn
on message tracking and check your tracking log. If it makes it to the
categorizer and gets the NDR, then it's global settings. If it doesn't make
it past the store, then it's store level mailbox limits. The categorizer is
responsible for enforcing sending and receiving message size. The store is
responsible for enforcing "prohibit send at" and "prohibit send and receive
at".
The message is to an internal recipient, so the SMTP virtual server and
Connector message size settings should not come into play.
