Yes you can.
If the servers and users are all on the same network then you could do
something like not give the server a default gateway, that would stop
mail going out.
Better would be to create an RG and SMTP connector. Use the address
space * and deny everyone from sending though it. That will result in
a message being sent back to the user saying they can't send to the
intended recipient.
Also make sure that the ISA does not accept SMTP (tcp 25) traffic from
within the network, that also prevents mail going out.
