Unfortunately in the interval we've switched from VS7.1 to
VS8.0, switched from x86 to x64 build machines, and in the process
upgraded our version of MDAC, so I'm not sure what might be causing
the problem.

Upgraded your version of MDAC on a x64 machine?
The normal versions of MDAC make it very clear that it is for Win32 _only_:

where it details various falvorus of Win32 under "Supported Operating
Systems" but not Win64.

and under no circumstances should you try to "upgrade" MDAC on a x64
machine.
The MDAC on the x64 is specific to x64 and should not be tampered with.
Only the security patches for MDAC that are marked x64 are applicable for
this.
x64 is a special case here.
The general MDAC is available for most of the Win32 family but not Win64.
It is an exception.

Can anyone suggest what might be the problem, or at least where to
start looking?

Reinstall Windows x64. I am 99% certain you cannot uninstall MDAC.
