Permission Denied message

I recently set up security levels using the security wizard on a database.
This is the first time I've done anything with security levels. I set myself
as the administrator and gave different levels of access to the various
users. All seemed to work well.
When I went to go back into the database I got the following message

'You do not have the necessary permissions to use the "...database name
,..." Have your system administrator or the person who created the object
establish the appropirate permissions for you"

I thought that I'd be asked for a username and password to access the
database but this came up before anything else. What have I done wrong in
setting things up.

(I did make a full copy of the database before I started fiddling, so all is
still running smoothly, but I do have to get these security levels in place)
